Abstract

In vitro regeneration protocol through the stem callus culture of Celastrus paniculatus Willd was established. The explants induced callus on LS medium supplemented with 2% fructose, 6 uM Kn and 1 uM IBA. Interaction of the same hormones at lower concentrations (1.5 to 3.5 uM Kn and 0.1 to 0.4 uM IBA) favoured shoot bud differentiation from the calli. The frequency of shoot bud production was optimized (30.4 shoots per callus) at the concentration of 2.5 uM Kn and 0.3 uM IBA. The microshoots rooted on LS half strength medium supplemented with 1.0 to 5.0 uM or 1.0 to 3.0 uM NAA. About 98% of the regenrants were successfully acclimatized to the natural environment. Electrophoretic analysis of isoenzyme and proteins of the regenerants exhibited similar banding patterns a compared to in vivo plant.
